Sendhil Ramamurthy nasce il 17 Maggio 1974 a Chicago, nell'Illinois. I suoi genitori sono entrambi medici e anche indiani. Ha sposato l'attrice Olga Sosnovska da cui ha avuto una bambina.
Inizialmente iscritto alla scuola di medicina per seguire le orme della propria famiglia, Sendhil, si è interessato durante il primo anno scolastico alla recitazione. Decide così di abbandonare gli studi per dedicarsi alla recitazione.
Si laurea nel 1999 presso la "Webber Douglas Academy of Dramatic Art". Il suo successo televisivo è arrivato con Heroes poichè ha convinto gli autori a tal punto che hanno riscritto la parte apposta per lui.
