Amaury Nolasco nasce a Puerto Rico. Inizialmente ha studiato biologia all'Università di Puerto Rico con l'intento di divenire dottore ma il destino ha voluto che un regista lo notasse e lo facesse partecipare ad uno spot. Amaury partecipa a numerose parti secondarie e dopodichè si trasferisce a New York dove partecipa all'American British Dramatic Arts School.
Nolasco studia molto e si specializza sulla dizione e la presenza scenica. Successivamente partecipa a molti tv show nel ruolo di guest star in "Arli$$", "CSI: Crime Scene Investigation" e "ER". Partecipa anche a "Orange Julius" e nel sequel di "Fast 2 Furious", "George Lopez", "Eve" e "CSI: NY".
