Frank Grillo nasce l'8 Giugno 1965 a New York. Inizia la sua strada di recitazione alle superiori ma i genitori insistono perchè lui finisca gli studi e così si laurea in Economia e Commercio alla New York University.
Inizia la sua carriera con alcune pubblicità come in "Silk Stalkings" e "Poltergeist: The Legacy" e successivamente prende parte alla serie TV "Sentieri". Ottiene alcuni ruoli anche in "Fiona", "Legacy" oltre a "Wasteland", "Battery Park", "For the People", "The Shield" e "Blind Justice".
All'inizio del 2006 partecipa anche a "Hollis & Rae", una serie Tv sfortunatamente non andata in onda.
