Wentworth Miller nasce il 2 giugno del 1972 a Chipping Norton, Inghilterra. Miller cresce a Brooklin, New York, per poi trasferirsi in Pennsylvania a Leetsdale ove si diploma alla Quaker Valley Senior High School.
Prosegue nella strada degli studi e si laurea all'università di Princeton in Letteratura Inglese. Intanto studia anche come cantante migliorando le sue doti artistiche presso i Princeton Tigertones, il gruppo musicale dell'università.
Dopo essersi laureato si reca a Los Angeles dove cerca di entrare nel mondo del cinema. Dopo qualche tempo ha ottenuto alcuni ruoli come in "Buffy the Vampire Slayer" e in "Popular". Successivamente ha partecipato ad un ruolo più importante interpretando le parti di David in "Dinotopia".
Nel 2003 debutta vel cinema col film "La macchia umana", presentato anche al Festival di Venezia 2003 insieme ad Anthony Hopkins e Nicole Kidman: Miller interpreta il protagonosita nell'età giovanile.
Ultimamente ha preso parte a "Underworld" e a "Joan of Arcadia".
