Eric Mabius è nato il 22 Aprile 1997 a Harrisburg in Pennsylvania. Mabius ricevette grandi elogi alla Amherst Regional High School tra cui il titolo come "miglior ragazzo drammatico". In seguito alla Amherst School si iscrive alla Sarah Lawrence College in Bronxville, dove studia cinema.

Nel 1995 Eric inizia la sua carriera recitando in "Resident Evil", "Chicago Hope", "Millennium", "Popular", "The O.C.", "Eyes" e in "The L Word".  

"Ugly Betty" è il suo grande successo del 2006 che gli fece guadagnare la nomina di uomo più sexy del mondo da People Magazine.
